V rámci projektu CasProt v spolupráci so skupinou Prof. Dr. Plückthuna z Univerzity v Zürichu sme v laboratóriách CIB vyvinuli matrix pre jednokrokovú izoláciu proteínov metódou afinitnej chromatografie. Základom tohto matrixu je proteín DARPin (Dizajnovaný Ankyrínový Repetitívny Proteín) s vysokou afinitou k maltózu-viažúcemu proteínu (MBP), ktorý sme pre upravili metódou racionálneho dizajnu pre potreby afinitnej chromatografie. V práci sme ukázali nielen vysokú afinitu takéhoto matrixu ku MBP konjugovaným proteínom, ale aj kapacitu a reprodukovateľnosť izolácie týmto spôsobom. Táto metóda bola opublikovaná v časopise International Journal of Biological Macromolecules (IF 6.853) (Nemergut M., Škrabana R., Berta M., Plückthun A., Sedlák E. Štruktúra komplexu DARPin-MBP, ktorý predstavuje základ vyvinutej purifikačnej metódy, s vyznačenými lyzínmi, ktoré boli modifikované racionálnym dizajnom (PDB: 1SVX a 1ANF).
